Loading ...ครั้งหนึ่งเราเคยทำให้ “เนื้อสมัน” สัตว์ที่มีเขาสวยงามที่สุดสูญพันธุ์ไปจากโลก และสัตว์ป่าอีกหลายชนิดกำลังลดจำนวนลงจนอยู่ในภาวะใกล้สูญสิ้นไปจากป่าเมือง ไทย แต่ความสำเร็จของนักวิจัยไทยในการทำ “ละมั่งหลอดแก้ว” ตัวแรกของโลกได้จุดความหวังในการคืนสัตว์หายากสู่ป่าอีกครั้ง
ทันทีที่เห็นหูเล็กๆ ของลูกละมั่งดุกดิกไปมาอยู่ใกล้ๆ ละมั่งตัวแม่ที่ได้รับการฝากตัวอ่อนจากการผสมแบบปฏิสนธินอกร่างกายหรือการ ผลิตตัวอ่อนหลอดแก้วหรือไอวีเอฟ (in vitro fertilization: IVF) น.สพ.สกนธ์ น้อยมูล สัตวแพทย์ผู้ดูแลละมั่งในโครงการผลิตละมั่งหลอดแก้วของส่วนอนุรักษ์วิจัยและ การศึกษา ขององค์การสวนสัตว์ในพระบรมราชูปถัมภ์ ตื่นเต้นจนทำอะไรไม่ถูก ได้แต่โทรแจ้งผู้ใหญ่และเจ้าหน้าที่ในโครงการ
เซอร์ไพรซ์! ละมั่งหลอดแก้วตัวแรกของโลก
ลูกละมั่งเพศเมียที่เกิดอยู่ในภายในสวนสัตว์เปิดเขาเขียวนี้คือสัตว์ ป่าที่เกิดด้วยวิธีผลิตตัวอ่อนหลอดแก้วตัวแรกของไทย และยังเป็นละมั่งหลอดแก้วตัวแรกของโลก โดย น.สพ.สกนธ์ บอกทีมข่าววิทยาศาสตร์ ASTV-ผู้จัดการออนไลน์ว่าเขามีส่วนร่วมในโครงการตั้งแต่การคัดเลือกแม่อุ้ม บุญ คอยกำจัดโรคที่เป็นสาเหตุให้แท้ง จนแข็งแรงดี ระหว่างท้องจะให้แคลเซียมและเกลือแร่เสริมเพื่อลดความเครียด ส่วนลูกละมั่งที่เกิดมาได้รับการดูแลเรื่องการถ่ายพยาธิเพียงอย่างเดียวก่อน และเมื่อครบปีจะไดั้บวัคซีนป้องกันบาดทะยัก ส่วนวัคซีนอื่นไม่จำเป็นเพราะยังไม่มีประวัติการติดโรคอื่นที่ต้องฉีดวัคซีน ป้องกัน
“หลังจากลูกละมั่งเกิดมาแล้วทีมสัตวแพทย์ต้องเฝ้าดูแลตลอด 24 ชั่วโมง และเฝ้าดูว่าแม่ละมั่งเลี้ยงลูกหรือไม่ เพราะมีปัจจัยเสี่ยงหลายอย่าง เป็นแม่อุ้มบุญด้วย ไม่ทราบว่าตัวเอง และมีสัญชาตญาณระวังภัย ทำให้อาจทิ้งลูก แต่แม่อุ้มบุญก็เลี้ยงลูกได้อย่างดี และลูกละมั่งแข็งแรงมาก เพราะได้ภูมิคุ้มกันจากนมแม่” น.สพ.สกนธ์กล่าว พร้อมบอกด้วยว่าลักษณะภายนอกของละมั่งท้องนั้นเหมือนละมั่งทั่วไป การมองด้วยตาเปล่าไม่สามารถบอกได้ นอกจากการเก็บมูลไปตรวจฮอร์โมนหาภาวะตั้งครรภ์ และตอนออกลูกละมั่งหลอดแก้วนี้แม่อุ้มบุญได้แอบไปคลอดอยู่ตรงพุ่มไม้
รวมสุดยอดทีมวิจัย
ความสำเร็จในการผลิตลูกละมั่งหลอดแก้วครั้งนี้ น.สพ.สุเมธ กมลนรนารถ หัวหน้าโครงการกล่าวว่ามีนักวิทยาศาสตร์ที่เกี่ยวข้องมากกว่า 30 คน โดยมีทั้งทีมคนเลี้ยงสัตว์ให้สมบูรณ์ ทีมรีดน้ำเชื้อ โดยพ่อพันธุ์ละมั่งชุดนี้เป็นพ่อพันธุ์จากสวนสัตว์ที่นครราชสีมาที่ขนย้าย โดยการแช่แข็งมาผสมพันธุ์กับแม่ละมั่งที่เขาเขียว ทีมสัตวแพทย์และผู้ช่วยสัตวแพทย์ นักวิทยาศาสตร์จากหลากหลายสาขาและระบบสนับสนุนต่างๆ อาทิ ฝ่ายโภชนาการ โรงพยาบาลสัตว์ป่า และผู้บริหาร เป็นต้น
น.สพ.สุเมธกล่าวว่ามีนักวิทยาศาสตร์ที่มีบทบาทสำคัญ 6 คน ซึ่งมี 2 คนเป็นชาวต่างชาติได้แก่ ดร.ปิแอร์ โคมิซโซลี (Pierre Comizzoli) นักวิจัยเชื้อสายฝรั่งเศส-อิตาลี จากสถาบันชีววิทยาการอนุรักษ์สมิทโซเนียน (Smithsonian Conservation Biology Institute) สหรัฐฯ ผู้เชี่ยวชาญด้านการเจาะไข่หรือการดูดไข่จากระบบสืบพันธุ์สัตว์เพศเมีย โดยเฉพาะสัตว์ตระกูลกวาง, ดร.เดบรา เบิร์ก (Dr.Debra Berg) นักวิจัยหญิงชาวอเมริกันจากสถาบันเอจีรีเสิร์ช (AG Research) นิวซีแลนด์ เธอเชี่ยวชาญเรื่องน้ำยาเพาะเลี้ยงในหลอดแก้วและการย้ายฝากตัวอ่อน โดยเฉพาะสัตว์ตระกูลกวางและกวางแดง (Red Deer) อีกทั้งยังทำการโคลนนิงแกะและแพะด้วย
นักวิทยาศาสตร์ไทยอีก 4 คนที่เหลือซึ่งมีบทบาทสำคัญในการผลติละมั่งหลอดแก้ว ได้แก่ ผศ.น.สพ.ธีรวัฒน์ ธาราศานิต จากคณะสัตวแพทย์ จุฬาลงกรณ์มหาวิทยาลัย ผู้ชำนาญด้านการแช่แข็งเซลล์สืบพันธุ์และการเลี้ยงเซลล์ไข่ไปปฏิสนธิในหลอด แก้ว ที่ผ่านมามีประสบการณ์ในการแช่แข็งตัวอ่อนสัตว์ตระกูลแมว ม้าและสุกร และผลิตลูกแมวหลอดแก้วมาแล้วหลายตัว, น.สพ.ดร.สิทธวีร์ ทองทพิย์ศิริเดช จากคณะสัตวแพทย์ มหาวิทยาลัยเกษตรศาสตร์ เชี่ยวชาญด้านการแช่แช็งอสุจิ โดยเคยเป็นหัวหน้าทีมวิจัยช้างและผสมเทียมช้างเชือกแรกได้สำเร็จคือ “พลายปฐมสมภพ” และมีส่วนร่วมในการผสมเทียมละมั่งสายพันธุ์พม่าจากอสุจิแช่แข็งได้ละมั่งเพศ ผู้ที่เกิดเมื่อ 14 ก.พ.52 ชื่อ “อั่งเปา”
สพ.ญ.ดร.อัมพิกา ทองภักดี ทีมงานสวนสัตว์เขาเขียว ผู้มีประสบการณ์โคลนนิงแมวลายหินอ่อน แมวป่าหัวแบน และกำลังวิจัยเรื่องระบบสืบพันธุ์แมวป่าอีกหลายชนิด โดยในการผลิตละมั่งหลอดแก้วนี้เธอมีส่วนในการผลิตตัวอ่อนละมั่งระยะ 7 วัน หรือในระยะบลาสโตซิสต์ (blastocyst) ซึ่ง น.สพ.สุเมธกล่าวว่า เธอประสานการทำงานร่วมกับ ดร.เดบราได้เป็นอย่างดี และ นสพ.ดร.บริพัตร ศิริอรุณรัตน์ หัวหน้าทีมวิจัยโครงการจากสวนสัตว์เปิดเขาเขียว ซึ่งมีประสบการณ์ทำงานจากสมิทโซเนียนและเป็นคู่หูของ ดร.โคมิซโซลี
ทำไมต้อง “ละมั่ง” ?
“ละมั่งมีในหลายประเทศ แต่ไทยเป็นประเทศเดียวที่มีละมั่งถึง 2 สายพันธุ์ คือสายพันธุ์ไทยและสายพันธุ์พม่า โดยสายพันธุ์ไทยจะพบทางฝั่งตะวันออกและสายพันธุ์พม่าจะพบทางฝั่งตะวันตกของ ประเทศ แต่น่าเสียดายที่สูญพันธุ์ไปจากธรรมชาติหมด ที่อินเดียมีละมั่งอินเดีย ที่พม่ามีละมั่งพม่า และที่ลาว เวียดนามและเขมรมีละมั่งไทย” ดร.สุเมธกล่าว และบอกว่าการผลิตละมั่งหลอดแก้วเป็นเรื่องยากมาก ซึ่งที่สถาบันสมิทโซเนียนของสหรัฐฯ มีความพยายามทำละมั่งหลอดแก้วสายพันธุ์ไทย แต่ยังไม่ประสบความสำเร็จในการฝากถ่ายตัวอ่อนในหลอดแก้วไปยังแม่อุ้มบุญ
เหตุผลที่เราต้องผลิตละมั่งหลอดแก้วนั้น นายภิมุข สิมะโรจน์ ผู้อำนวยการองค์การสวนสัตว์ ในพระบรมราชูปถัมภ์ กล่าวว่าครั้งหนึ่งเราเคยมีสมันแต่สูญพันธุ์ไปหมดไม่เหลือ จึงไม่อยากให้เป็นเช่นนั้นอีกในละมั่ง ซึ่งเป็น 1 ใน 15 สัตว์สงวนของไทย และไม่เหลือในธรรมชาติมา 50 ปีแล้ว แต่ข้อจำกัดในการขยายพันธุ์ละมั่งตามวิธีธรรมชาติคือได้ต้นพันธุ์ที่มีปัญหา การผสมเลือดชิด (inbreeding) ทำให้ได้ละมั่งที่ไม่แข็งแรง จึงต้องคิดเทคโนโลยีเพื่อหาสายพันธุ์ที่สมบูรณ์ และเมื่อวันที่ 30 มี.ค.53 ได้มีการประชุมเชิงปฏิบัติการเรื่องละมั่งเป็นครั้งแรก จนกระทั่งเมื่อวันที่ 21 ก.พ.54 ได้ย้ายฝากตัวอ่อน และเมื่อวันที่ 17 ต.ค.54 จึงได้ละมั่งหลอดแก้วตัวแรกของโลก
ตัวอ่อนหลอดแก้วแก้ปัญหาเลือดชิด
การผลิตละมั่งหลอดแก้วแก้ปัญหาเลือดชิดได้อย่างนั้น ดร.น.สพ.บริพัตร หัวหน้าทีมวิจัยอธิบายว่า หากปล่อยให้ละมั่งขยายพันธุ์ตามธรรมชาติ ตัวผู้และตัวเมียที่เป็นพี่น้องกันนั้นมีโอกาสผสมพันธุ์กันได้ง่าย ทำให้ได้ลูกที่เลือดชิด อาจทะให้มีอาการปากแหว่ง ตาบอดหรือตายได้ ดังนั้น จึงต้องทำให้ความหลากหลายทางพันธุกรรมด้วยการผลิตตัวอ่อนแบบหลอดแก้ว ซึ่งเราทราบว่าน้ำเชื้อตัวผู้นั้นมาจากไหน และการใช้ฮอร์โมนกระตุ้นจะทำให้ได้ไข่มากถึง 20 ใบ ซึ่งนำไปผสมกับเชื้อตัวผู้และคัดเลือกพันธุ์ที่ดีเพื่อขยายต่อไป
“ส่วนเหตุผลที่ต้องฝากตัวอ่อนให้แม่อุ้มบุญนั้น เพราะแม่อุ้มบุญไม่จำเป็นต้องมีสายพันธุ์ดี โดยเลือกตัวที่มีพี่น้องเยอะซึ่งแสดงว่าเป็นเลือดชิดเยอะแล้ว ควรหยุดได้แล้ว จึงเอามาเป็นโรงงานผลิตลูกแทน” ดร.น.สพ.บริพัตรให้เหตุผล นอกจากนี้ ผศ.น.สพ.ดร.ธีรวัฒน์ ยังได้อธิบายเพิ่มเติมแก่ทีมข่าววิทยาศาสตร์ว่า หากให้ละมั่งที่เป็นแม่พันธุ์ทำหน้าที่ตั้งท้องก็จะขยายพันธุ์ได้ครั้งละ 1 ตัว แต่ถ้าใช้แม่อุ้มบุญจะทำให้ขยายพันธุ์ได้ครั้งละหลายสิบตัว
สำหรับละมั่งหลอดแก้วตัวแรกของโลกนี้ทางทีมวิจัยได้เตรียมทูลเกล้าฯ ถวายแด่พระบาทสมเด็จพระเจ้าอยู่หัว และเมื่อพิสูจน์แล้วว่าเทคโนโลยีในการขยายพันธุ์นี้ได้ผลดี ก็จะผลิตละมั่งหลอดแก้วเพื่อปล่อยคืนสู่ธรรมชาติต่อไป แต่การปล่อยคืนสู่ป่านั้นเป็นเรื่องซับซ้อน โดย น.สพ.ดร.บริพัตรอธิบายว่าเราปล่อยละมั่งเพื่อไปเป็นเหยื่อ ต้องไปอยู่รวมกับเสือ งู หมาไนและนักล่าอื่นๆ จึงเป็นเรื่องซับซ้อน สถานที่ปล่อยนั้นต้องเป็นป่าที่เหมาะสม มีอาหาร มีพื้นที่ให้วิ่งหนีนักล่า โดยก่อนหน้านี้ทางองค์การสวนสัตว์ฯ ได้ร่วมกับมหาวิทยาลัยเกษตรศาสตร์และกรมอุทยานแห่งชาติ สัตว์ป่าและพันธุ์พืชปล่อยละมั่ง 70 ตัวสู่ป่าห้วยขาแข้ง และปัจจุบันเหลือประมาณ 10 ตัว
พลาดขั้นตอนเดียว…ทุกอย่างจบ
ผศ.น.สพ.ดร.ธีรวัฒน์กล่าวว่าการนำเทคโนโลยีต่างๆ มาใช้กับสัตว์ป่าไม่ง่าย อย่างแรกคือไม่มีข้อมูลเลยจึงต้องอาศัยผู้เชี่ยวชาญจากหลายด้าน โดยเลือกเทคโนโลยีที่มีโอกาสประสบความสำเร็จสูงมาใช้ใช้วิทยาการจากหลายภาค ส่วนที่สามารถผลิตลูกสัตว์มาใช้ รวมถึงใช้เทคโนโลยีที่ผ่านการพัฒนาในสัตว์หลายชนิดมาแล้ว ซึ่งแต่ละขั้นมีความสำคัญเท่ากันหมด เพราะหากมีขั้นตอนหนึ่งขั้นตอนใดทำไม่ได้ทุกอย่างก็จบ
กว่าจะได้ละมั่งหลอดแก้วตัวแรกนี้ทีมวิจัยเคยล้มเหลวครั้งหนึ่ง คือเมื่อวันที่ 30 มี.ค.53 ทีมวิจัยได้ย้ายฝากตัวอ่อนสู่ท่อนำไข่ของแม่ละมั่งอุ่มบุญ 3 ตัวเป็นครั้งแรก ณ สวนสัตว์เขาเขียว และพบว่าแม่อุ้มบุญ 2 ตัวตั้งท้อง ซึ่งคิดเป็นความสำเร็จ 66.7% แต่ละมั่งทั้งสองได้คลอดลูกตายก่อนกำหนดเมื่อมีอายุการตั้งท้องประมาณ 7 เดือน จากนั้นจึงมีความพยายามในครั้งที่ 2 คือเมื่อเดือน ก.พ.54นี้ ซึ่งได้ย้ายฝากตัวอ่อนสุ่แม่อุ้มบุญ 8 ตัว และพบว่าแม่อุ้มบุญ 1 ตัวตั้งท้อง
สอดคล้องกับคำอธิบายของ น.สพ.ดร.บริพัตรที่แจกแจงว่าแต่ละขั้นตอนนั้นมีความสำคัญและมีโอกาสล้มเหลว ทุกขั้นตอน ตั้งแต่การแช่แข็งเซลล์ทั้งไข่และอสุจิที่มีโอกาสตายและฝ่อได้ เมื่อปฏิสนธิแล้วก็ตายได้ หรือเมื่อย้ายฝากไปยังแม่อุ้มบุญก็มีโอกาสไม่สำเร็จสูงมาก เมื่อทำสำเร็จแล้วจึงอยากส่งต่อความภูมิใจไปยังคนไทยทุกคน นอกจากนี้แล้วทางทีมวิจัยยังมีห้องปฏิบัติการผลิตตัวอ่อนที่มีมาตรฐานเทียบ เท่าคลีนิคผู้มีบุตรยากของโรงพยาบาลเอกชนที่มีชื่อเสียงใน อ.ศรีราชา จ.ชลบุรี
“เราคุยกันเรื่องละมั่งมาเป็น 10 ปี เรื่องจากการผสมเทียม มาจนถึงไอวีเอฟ วันหนึ่งๆ เราผ่าตัด (ละมั่ง) กันเป็น 10 ตัว ไม่เคยมีใครในโลกทำได้มาก่อน ภูมิใจในงานนี้มาก” น.สพ.ดร.สิทธวีร์ ผู้มีบทบาทในการแช่แข็งน้ำเชื้อกล่าว
ทางด้าน สพ.ญ.ดร.อัมพิกา ความสำเร็จในครั้งนี้เป็นบทพิสูจน์ว่าเทคโนโลยีการสืบพันธุ์ในสัตว์เลี้ยง นั้นนำมาใช้ในสัตว์ป่าได้จริงๆ และเธอโชคดีที่ได้รับการผลักดันให้ศึกษามาในเรื่องนี้ซึ่งคิดว่าเทคโนโลยี นี้จะเป็นหนทางในการอนุรักษ์ความหลากหลายทางพันธุกรรมและอนุรักษ์สัตว์ป่า ได้ ส่วนก้าวต่อไปคือการพัฒนาเทคโนโลยีและสร้างธนาคารพันธุกรรมเพื่อผลิตสัตว์ พันธุ์ดีและผลิตสัตวแพทย์รุ่นใหม่ และอนาคตจะได้ก้าวต่อไปสู่การขยายพันธุ์สัตว์ชนิดอื่นๆ
สำหรับละมั่งหลอดแก้วในโครงการนี้เป็นละมั่งสายพันธุ์พม่า ซึ่งมีอยู่ในแหล่งเพาะเลี้ยงเมืองไทยประมาณ 1,000 ตัว ส่วนสายพันธุ์ไทยเหลืออยู่ในสถานที่เลี้ยงประมาณ 40 ตัว โดนทีมวิจัยไม่ทราบว่ายังมีอยู่ในสถานเลี้ยงสัตว์เอกชนอีกหรือไม่ ด้วยจำนวนละมั่งพม่าที่มีเยอะกว่านี่เองทีมวิจัยจึงไม่อยากเสี่ยงทำวิจัยใน ละมั่งสายพันธุ์ไทย เมื่อได้ผลการทดลองที่ดีในสายพันธุ์พม่าแล้วจึงจะขยายสู่สายพันธุ์ไทยต่อไป
